Many of my contacts saved in address book in Thunderbird have disappeared and also the memory for typing existing email addresses into the email address line does not work so where I'd usually type the first few letters of an email and it gives me the rest of the email address this no longer happens.

re ;also the memory for typing existing email addresses

There is no memory - it uses the address book, so if contacts are not in address book then they will not appear in the drop down. This explains why they are not appearing in the drop down autofill section.

Create a backup of your address book so that you do not lose any more contacts. In Address Book select the address book Tools > Export Save in a suitable location as either LDIF or comme separated.csv file.
